HILIC概念最早由Andrew Alpert于1990年提出,旨在解决传统反相色谱(RPLC)对强极性化合物(如糖类、肽类、核酸等)分离效率低的问题。早期采用高比例有机相(如乙腈/水)作为流动相,结合硅胶或氨基键合固定相,通过亲水相互作用实现分离。随着新型固定相材料(如两性离子硅胶、聚合物基质)和仪器技术(如高精度泵、质谱联用)的发展,HILIC方法的重复性和适用范围显著提升。HILIC与高分辨质谱(HRMS)联用成为主流,推动其在复杂生物样本(如血浆、尿液)和外源性物(如药物代谢物、环境污染物)分析中的应用。如今,HILIC已应用广泛于生物样本分析、临床诊断、天然产物与药物开发、食品与食品安全、环境监测和制药工业

iHILIC®-(P) Classic 色谱柱
iHILIC色谱柱专为通过亲水作用液相色谱法(HILIC)分离和纯化极性及亲水性化合物而设计。其填充固定相采用两性离子型电荷修饰的酰胺硅胶或聚合物颗粒,这些基质通过共价键合中性、正电荷及负电荷的亲水功能基团。因此,iHILIC色谱柱的分离机制基于亲水作用、氢键作用和弱静电相互作用的协同效应,具有独特的分离选择性和高分离效率。
iHILIC色谱柱提供四种不同的表面化学特性,可提供互补的选择性以满足多样化分析需求。
